Kit Components:
Each kit includes the Label IT® miRNA Cy™3 and Cy™5 Reagents, Reconstitution Solution, 10X Labeling Buffer M, 10X Stop Solution, Purification Columns (with Collection Tubes), Column Binding Buffer, Column Wash Buffer, Column Elution Solution, and Elution Tubes. This kit also contains a 2X Hybridization Solution.

Figure 1. The Label IT® miRNA Labeling Kits. The Label IT Cy™3, Cy™5, and biotin reagents are composed of three regions: the Cy3, Cy5 or biotin label (green), the linker (yellow) which facilitates electrostatic interactions with nucleic acids and the reactive alkylating group (blue) that covalently attaches the Label IT reagents to any reactive heteroatom within miRNAs. The covalent attachment of the Label IT reagents to miRNA does not alter the structure or downstream hybridization performance of the miRNA, and thus, once labeled, the miRNA samples can be hybridized to miRNA-specific microarrays and analyzed to determine the miRNA expression profile.

Figure 2. The Label IT® miRNA Labeling Kit is More Sensitive Than NCode™. A pool of 699 synthetic miRNAs was labeled and hybridized to the NCode™ Multi-Species miRNA Microarray V2 at varying input amounts. Features with signal greater than 3X background were considered detected.
